Production of Universal Red Blood Cells and Platelets in a Novel Bioreactor |

| National Institute of Standards and Technology/ATP The goal of the proposed project is the cost-effective ex vivo production of human red blood cells (RBCs) and platelets from hematopoietic stem cells (HSCs). We will use our patented, biocompatible, porous three-dimensional tissue scaffold, termed Cellfoam(TM) (U.S. Patent 5,282,861), to culture human HSCs in a continuous perfusion system to mass produce safe, universally compatible RBCs and platelets. Cellfoam enables the survival and expansion of normal human HSCs in a manner and to an extent that is not possible with other devices. In preliminary investigations, we have demonstrated that Cellfoam enables the long-term culture of HSC in the absence of supplemented cytokines, expands HSCs, and maintains the multipotent ability of HSCs to make RBC, WBC and platelet progeny. The continuous perfusion Cellfoam system will be developed through the following three specific aims: 1) Development of Cellfoam capabilities for continuous HSC culture 2) Development and integration of our continuous perfusion system prototype 3) Red blood cell and platelet production from the Cellfoam perfusion system |
